Genetic analysis of Murine hepatitis virus nsp4 in virus replication.
Coronavirus replicase polyproteins are translated from the genomic positive-strand RNA and are proteolytically processed by three viral proteases to yield 16 mature nonstructural proteins (nsp1 to nsp16). nsp4 contains four predicted transmembrane-spanning regions (TM1, -2, -3, and -4), demonstrates characteristics of an integral membrane protein, and is thought to be essential for the formatio...
متن کاملProtease bypass of temperature-sensitive murine leukemia virus maturation mutants.
Cells infected with certain temperature-sensitive mutants of Moloney murine leukemia virus synthesize the virion precursor proteins but neither bud virions nor cleave precursor proteins to their mature form. Addition of proteases to cells infected with these mutants caused cleavage of the precursor proteins Pr65gag and Pr180gag-pol to their mature forms at the nonpermissive temperature. Concomi...
متن کاملIsolation of temperature-sensitive mutants of murine sarcoma virus.
Three separate murine sarcoma virus nonproducer cell lines have been isolated which are temperature sensitive for the maintenance of transformation. In each case, a viral rather than a cellular genetic mutation is the reason for the temperature-sensitive effect. Superinfection of one of the mutants with murine leukemia virus overcomes the temperature-sensitive change in the transformed state.
متن کاملMapping of RNA- temperature-sensitive mutants of Sindbis virus: complementation group F mutants have lesions in nsP4.
Temperature-sensitive (ts) mutants of Sindbis virus belonging to complementation group F, ts6, ts110, and ts118, are defective in RNA synthesis at the nonpermissive temperature. cDNA clones of these group F mutants, as well as of ts+ revertants, have been constructed. To assign the ts phenotype to a specific region in the viral genome, restriction fragments from the mutant cDNA clones were used...

were isolated after mutagenesis by 5-azacytidine, 5-fluorouracil, or proflavine. The restricted replication of all mutants at 39 C was blocked subsequent to cell penetration and could not be attributed to heat inactivation of virus infectivity. Complementation analysis was made possible through the use of poly-L-ornithine. The members of one complementation group exhibited wild-type RNA synthes...
